Project description:

Recent mouse studies have documented a pivotal growth-regulatory role of the Cullin7 (CUL7) E3 ubiquitin ligase (1,2). In addition, CUL7 germline mutations were linked to 3M-syndrome, a hereditary disorder characterized by pre- and postnatal growth retardation in humans (3). We have identified Insulin Receptor Substrate-1 (IRS-1), a critical mediator of the insulin / insulin-like growth factor 1 (IGF-1) signaling pathway, as a proteolytic target of the CUL7 E3 ligase (4). CUL7-/- cells were found to accumulate IRS-1 and exhibit increased activation of pro-mitogenic IGF-1 pathway. However, CUL7-/- cells grew poorly and displayed features of oncogene-induced senescence (OIS), a tumorsuppressive mechanism of the cell in response to sustained oncogenic signaling (5).

The aim of this project is to study the role of the CUL7 E3 ligase for cellular senescence, to identify underlying signaling pathways and to characterize their pathomechanistic relevance for CUL7 linked growth retardation syndromes.The project will cover a broad range of state-of-the-art methods of cell biology and ubiquitin biochemistry. Salary and contract according to German TVöD.  

Requirements and application: 

The ideal candidate has a PhD degree in molecular cell biology, biochemistry or a closely related field and a background in ubiquitin biology.
