Netherlands : PhD student for Chromosome Architecture during Early Development

Responsibilities
The development of a single cell into a complex, multicellular organism is one of the most fascinating topics in biology. Essential for pluripotency and differentiation is the cell's state of chromatin, and how genes are marked for activation or repression by epigenetic mechanisms. The embryonic transcription machinery accommodates the requirements of development using a diverse array of molecular mechanisms. Little is known, however, about how enhancers and other regulatory elements are marked for activity and how this relates to chromosome folding during the early stages of pluripotency and lineage commitment. We have discovered that key histone modifications are deposited de novo in a stepwise fashion on unmarked chromatin, revealing a hierarchy of activation and repression. The results provide many tangible opportunities to dissect molecular mechanisms of epigenetic regulation during the very dynamic stages of early development.

Work environment
Gene regulation is a central theme in the research of the Department of Molecular Biology (Faculty of Science, Radboud University Nijmegen), with particular emphasis on genomics, proteomics and analysis of the transcriptome. Genetic information is selectively expressed, for example, in cell type-specific gene regulation during embryonic development. A major aspect is the application of fundamental knowledge of gene regulation in the prevention and cure of cancer and infectious diseases. The department is part of the Nijmegen Centre for Molecular Life Sciences (NCMLS), a joint research institute of the Radboud University Nijmegen Medical Centre and the Faculty of Science. NCMLS focuses on basic science and its translation into pioneering treatments for disease

What we expect from you
You should be a biologist or biochemist with a strong interest in gene regulation and embryonic development, and keen to participate in an interesting and innovative research project in genomics, epigenetics and gene regulation using Next Generation Sequencing approaches. You should have a M.Sc. degree in (Medical) Biology, Molecular Life Sciences or Biochemistry, a strong motivation to conduct scientific research, affinity for genome-wide approaches, and preferably some background in the areas of transcription, genomics, epigenetics or development.

What we have to offer
We offer you:
- employment: 1,0 fte;
- a maximum gross monthly salary of € 2,612 based on a 38-hour working week;
- in addition to the salary: an 8% holiday allowance and an 8.3% end-of-year bonus;
- the gross starting salary amounts to €2,042 per month based on full-time employment, and will increase to €2,612 per month in the fourth year;
- duration of the contract: 4 years;
- you will be appointed for an initial period of 18 months, after which your performance will be evaluated. If the evaluation is positive, the contract will be extended by 2.5 years.
